Molecular Cloning and Characterization of the Phenylalanine Aminomutase Gene From Taxus baccata L.
Authors
Abstract:
Background: Taxol is one of the most important anti-cancer drugs, which is obtained from yew trees (Taxus sp.). T+he fist step in side chain assembly of taxol is catalyzed by phenylalanine aminomutase, which converts α-phenylalanine to β-phenylalanine. Objectives: In this study, for the fist time, we report on the cloning, preliminary expression and characterization of a full-length gene and cDNA encoding phenylalanine aminomutase from Taxus baccata L. Materials and Methods: Comparison of the full-length gene with other ones identifid from the Taxus species showed high similarity, particularly with Taxus x media. Results: The results showed that the expression level of this gene in Taxus baccata is very low and therefore this enzymatic step could be a rate limiting step in the taxol biosynthesis pathway. Successful amplifiation of the cDNA was only obtained from RNA samples isolated from methyl jasmonate elicited suspension cells of Taxus baccata. The cloned cDNA contained a 2064 bp open reading frame encoding a protein composed of 687 amino acids. Sequence comparison analysis revealed that the gene is very similar (98 - 99 %) with respect to the nucleotide and amino acid sequences in diffrent Taxus species and also share the signature active site motif (175ASG177). Conclusions: The predicted structure of TbPAM was analyzed using bioinformatic tools. The results indicated that the protein has similar overall folding to tyrosine aminomutase.

full textAnti-ulcerogenic lignans from Taxus baccata L.
Four lignan type compounds, lariciresinol, taxiresinol, isolariciresinol and 3-demethyl-iso-lariciresinol, were isolated from the heartwood of Taxus baccata L. (Taxaceae) growing in Turkey through chromatographic techniques. In vivo anti-ulcerogenic potency of these compounds was investigated on ethanol-induced ulcerogenesis model in rats at two different doses, 50 and 100 mg/kg. All compounds ...

Three lignan derivatives, (-)-taxiresinol (1), (-)-3'-demethylisolariciresinol-9'-hydroxyisopropylether (2) and (-)-3-demethylisolariciresinol (3), previously isolated from the heart-wood of Taxus baccata L. (Taxaceae) were investigated for cytotoxicity against the Oncology Cell Line Panel (breast, colon, ovary, prostate, lung and a normal adult bovine aortic endothelial cell line) as well as f...
full textMolecular cloning and characterization of the yew gene encoding squalene synthase from Taxus cuspidata.
The enzyme squalene synthase (EC 2.5.1.21) catalyzes a reductive dimerization of two farnesyl diphosphate (FPP) molecules into squalene, a key precursor for the sterol and triterpene biosynthesis. A full-length cDNA encoding squalene synthase (designated as TcSqS) was isolated from Taxus cuspidata, a kind of important medicinal plants producing potent anti-cancer drug, taxol. The full-length cD...
full textMolecular and Morphological Characterization of a Taxol-Producing Endophytic Fungus, Gliocladium sp., from Taxus baccata
The endophytic fungal populations of different tissues of Taxus baccata grown at high altitudes in West Bengal, India were explored. These isolated fungal populations represented different genera, which were screened for taxol production using immunoassay technique. The culture AAT-TS-4(1) that produced taxol was identified as Gliocladium sp. based on its cultural, morphological characteristics...
full textMolecular cloning, characterization and expression of the phenylalanine ammonia-lyase gene from Juglans regia.
Phenylalanine ammonia-lyase (PAL) is the first key enzyme of the phenypropanoid pathway. A full-length cDNA of PAL gene was isolated from Juglans regia for the first time, and designated as JrPAL. The full-length cDNA of the JrPAL gene contained a 1935bp open reading frame encoding a 645-amino-acid protein with a calculated molecular weight of about 70.4 kD and isoelectric point (pI) of 6.7. Th...
